Peptide purity testing, explained
"99% pure" means nothing without proof. This guide explains how peptide purity is actually measured, what a certificate of analysis contains, and the checks worth making before you order from anyone.
How purity is measured
The industry-standard method is high-performance liquid chromatography (HPLC), which separates a sample into its components and reports what fraction is the target compound. Mass spectrometry (MS) complements it by confirming the compound's molecular identity — the mass has to match the sequence being sold.
A purity number without identity confirmation is incomplete: HPLC can show a clean single peak for the wrong compound.
Reading a certificate of analysis
A usable COA states the compound name and sequence, the testing laboratory, the test date, the batch number, and the purity result — ideally with the chromatogram attached. It should be recent (the current batch, not a supplier's best batch from two years ago) and from a laboratory that does not work for the seller.
Red flags: no batch number, a stock image instead of a dated report, a missing laboratory name, or a COA for a different product than the one for sale.
Why it matters for research
Unknown impurities make results uninterpretable: if a study sample contains fillers, truncated sequences, or a different molecule entirely, whatever happens in the experiment cannot be attributed to the peptide under investigation. Purity verification is not paperwork — it is what makes research data mean anything.
